Semaglutide

Also known as: Ozempic, Wegovy, Rybelsus

Fat Loss / GLP-1Grey areaHalf-life: 7d

Overview

Semaglutide is a GLP-1 receptor agonist approved for type 2 diabetes and chronic weight management. Clinical trials show average weight loss of 15-17% body weight. It is currently one of the most widely prescribed weight loss medications globally.

How it works

Mimics endogenous GLP-1 by binding to GLP-1 receptors in the pancreas, brain, and gut. Increases insulin secretion in a glucose-dependent manner, suppresses glucagon, slows gastric emptying, and acts centrally to reduce appetite and food intake.

Primary uses

  • Fat loss
  • Type 2 diabetes
  • Appetite suppression
  • Cardiovascular risk reduction
  • Blood sugar regulation

    • Appetite suppression begins within days of the first dose
    • Some nausea is common early on — typically resolves within 2-4 weeks
  2. Days 14–56

    • Consistent appetite reduction and slower gastric emptying are established
    • Weight loss of 1-3kg is typical in the first month
    • Blood sugar control is measurably improving
  3. Days 56–168

    • Average weight loss of 10-15% is typical at 3-6 months
    • Cardiovascular benefit markers are improving
    • Dose escalation may be appropriate — review with your provider
  4. Days 168–999

    • Long-term cardiovascular protection is one of semaglutide's strongest benefits
    • Sustained weight loss maintenance requires continued use
    • Consider reviewing your dose and goals with your provider
